Growth and sporulation of Hirsutella thompsonii in natural media and the conservation of its viability in different supports

The hyphomycete acarine pathogen, Hirsutella thompsonii, was
grown in liquid culture media composed of natural carbon and
nitr ogen sources cointained in wheat bran , potato-carrot, corn
meal and corn steep liquor , with or without 0 . 5 ml of Tween 80 and
incubated for 13 days at 26-28°C in a s haker bath at 70 hits;min.
Fungal biomass was determined by the mycelium dry weight, and
sporulation by number of conidia/ml. Biomass production was higher
in wheat bran, and in corn meal media with Tween 80. The higher
concentration of conidia ( 5.5 x 104 conidiafml ) was observed in
wheat bran medium without Tween 80 . The addition of Tween 80 did
not increase the production of conidia. The fungus was stored in
straw, wheat bran, sterile distilled water, physiological saline
solution, mineral oil or 10% glicerol, and in anhydrous silica gel,
at room and refr igeration temperatures . Low temperature was more
efficient in maintaining viability during the storage period
