Isolation and regeneration of Rhynchosporium secalis protoplasts the causal agent of barley scald

Protoplasts from conidia and mycelium were isolated from the fungus Rhynchosporium secails by using Novozymc
234 at 10 mg/ml. The age of the culture was a determinant factor to obtain protoplasts from mycelium. but it did
not have a significant effect in the case of conidia. Shaking during incubation of the fungus with I be enzy1nc was
imponant in the production of protoplasts from mycelium, while it was not important for protoplast isolation from
conidia. Fluorescent microscopy revealed that the protoplasts were mainly uninuclcatcd 90-100% when obtained
from conidia. and 70 -80% uninucleated when obtained from mycelium. The remaining protoplasts bearcd two
nuclei. Ten percent of the total protoplasts were regenerated and gave rise to discrele colonies on the medium used.
